1. LB膜界面修饰与伏安传感电化学应用研究
LB膜技术可以在分子水平上制造出均匀、超薄、厚度和分子层数可控的分子组合体修饰薄膜。在这种薄膜上,修饰成分具有排列紧密有序,活性中心密度较大,电子或物质传输容易等特点;而且,LB膜较牢固,所构筑的伏安传感器具有稳定性好、响应灵敏度高、使用较长的优势。LB膜的上述特点使其在电分析化学领域取得了一定程度的进展。我们的研究是将杯芳烃、DNA、碳纳米管、石墨烯等具有特殊功能的分子(材料)利用LB膜技术构筑伏安传感器,其结合LB膜技术与功能分子材料两者的优势,解决电极界面上一些科学问题。
2. DNA分子与靶向分子相互作用的电化学研究
DNA是许多抗癌药物作用的靶分子。DNA与其靶向分子相互作用的研究在探讨生命过程机理、DNA生物传感器的研制、药物的设计与合成以及药物抗病机理的研究等方面都具有重要意义。几十年以来,人们已经从分子水平上证实:药物分子是通过插入(Intercalation)、沟槽键联(Groove binding) 或共价键联(Covalent bonding) 三种基本方式与DNA结合在一起,并经常凭借氢键(Hydrogen binding)和库仑力(Coulombic force)使相互作用加强。大多数抗癌药物通过插入或沟槽键联与DNA相互作用;与DNA共价键联的抗癌药物较少。DNA与药物分子的非共价结合相互作用主要有三种模式:(1)外源分子插入DNA分子双螺旋的碱基之间;(2)外源分子与DNA分子的带负电荷的核糖-磷酸骨架之间的静电相互作用;(3)靶向分子与DNA的大沟或小沟的碱基对边缘直接发生相互作用。我们的研究着眼于抗癌中草药与DNA相互作用的方式、方法和作用参数,研究手段以电化学技术为主。该研究为抗癌中草药的开发、抗癌机理和临床试验提供基础理论数据。
3. 生物分子电化学研究
大脑神经中枢的活动机理历来是生命科学研究的重大课题,人们一直期待着彻底揭示神经活动的奥秘,并在当前高新技术、医学、药学等领域中加以应用。近年来的研究表明,整个中枢神经系统中有多种化学物质参与和影响着人类和动物复杂的神经活动,如脑神经传导过程由神经末梢释放出化学物质并进入突触间隙。由于这些化合物直接影响人体的行为和活动,参与体内的循环和调控,并与多种功能性疾病(如精神分裂症、忧郁症等)和器质性病变(如巴金森氏综合症等)息息相关,因此,研究神经传导物质-神经递质的电子传递机理、建立快速、灵敏、可靠的检测方法,在体、实时地定量测定这类神经活动物质具有重大意义。
